New paper in Trends in Cell Biology : Atomic force microscopy (AFM) allows researchers to image and manipulate cells at single-molecule resolution. In this issue, Müller and Dufrêne describe the basic principles of AFM, the advantages and limitations of the technique, and its recent application in investigating cell biological properties of the cell surface. • 14 June 2011. New book just published "Life at the Nanoscale: Atomic Force Microscopy of Live Cells". Life at the Nanoscale provides a state-of-the-art overview about how atomic force microscopy (AFM) can be used to image living cells, to probe their attachment to substrates or to other cells, to measure their mechanical properties, and to assess the forces in single-molecule interactions. Although much is known about the structure and biosynthesis of teichoic acids (TAs), details of their spatial organization are poorly understood. Using a powerful combination of single-molecule atomic force microscopy and fluorescence microscopy, Andre et al. define the distribution of TAs in relation with their physiological roles (Highlighted in the editorial & in Biopolymers). • 14 March 2011. She will be developing advanced force nanoscopy tools for understanding the molecular mechanisms of microbial cell adhesion. • 18 February 2011. Congratulations to David Alsteens for successful completion of his Ph.D. thesis entitled "Nanomechanics of the yeast cell wall". To date, nearly all single-molecule force measurements have been conducted in vitro where the system under study can be tightly controlled. Müller and colleagues argue that despite the extreme challenges involved, it is imperative that more of these experiments be moved into living cells where the biological molecules under investigation can operate under native conditions. They discuss five challenges that must be overcome to make this important move. • 16 December 2010. Congratulations to David Alsteens and to our collaborator Prof. Lipke (Brooklyn College of City University of New York) for their publication in the Proc. Natl. Acad. Sci. USA entitled "Force-induced formation and propagation of adhesion nanodomains in living fungal cells". Alsteens et al. use single-molecule atomic force microscopy to demonstrate that mechanical stimuli can trigger adhesion nanodomains in fungal cells. The results suggest that the force-induced clustering of adhesins may be a general mechanism for activating cell adhesion in microbial pathogens and open up exicting prospects for fighting fungal infections via the design of anti-adhesion molecules (Highlights in PNAS, Le Soir • 12 October 2010. Research Highlight in Nature: Imaging - Cell wall secrets. Yves Dufrêne at the Catholic University of Louvain in Belgium and his colleagues used atomic force microscopy to capture nanometre-scale images of the surface of Lactococcus lactis cells. The researchers say that their method could be used to help visualize peptidoglycan–drug interactions in other bacteria. • 15 June 2010. Congratulations to Guillaume Andre and to our collaborators Prof. Pascal Hols (ISV-UCL) and Dr. Saulius Kulakauskas (INRA) for their publication in Nat. Commun. “Imaging the nanoscale organization of peptidoglycan in living Lactococcus lactis cells”. The spatial organization of petidoglycan, the major constituent of bacterial cell walls, is an important but unsolved issue. Andre et al use atomic force microscopy to reveal the underlying cell-wall structure of mutant Lactococcus lactis. The images show dividing bacteria with periodic peptidoglycan bands running parallel to the short axis of the cell. • 14 June 2010. Together with our collaborator Prof. Jürgen Heinisch, we publish a paper in PLoS ONE entitled "Single-molecule atomic force microscopy reveals clustering of the yeast plasma-membrane sensor Wsc1". The lateral organization of the plasma membrane in microdomains is an important, yet largely unsolved problem in cell biology. Here we demonstrate the ability of AFM to resolve the clustering of a transmembrane protein, the cell integrity sensor Wsc1, in living Saccharomyces cerevisiae cells. • 26 October 2009. Research Highlight in Nature Reviews Microbiology, Fungal physiology: Stressed fungi are not highly sprung. Saccharomyces cerevisiae maintains cellular integrity in the presence of cell wall stress by upregulating cell wall-remodelling pathways. Such stresses are detected by a group of five transmembrane sensor proteins, Wsc1–Wsc3, Mid2 and Mtl2. Dupres et al. used atomic force microscopy (AFM) to show that Wsc1 behaves like a nanospring that is capable of resisting high mechanical force in vivo. • 9 October 2009. Science editors'choice, Biophysics: Embedded Sensors. Drugs used in the treatment of fungal infections are often directed against the protective fungal cell wall. Hence, a better understanding of how cell wall integrity is maintained under dynamic and stressful conditions has pharmaceutical implications. Dupres et al. have used atomic force microscopy (AFM) to characterize the response of the yeast plasma membrane protein Wsc1 to mechanical stress. • 10 December 2008. The Dufrêne lab studies the nanoscale surface architecture, properties and interactions of microbial cells, including pathogens, using atomic force microscopy (AFM) techniques. The strategy involves developing advanced AFM methods (cell preparation and tip functionalization, high-resolution imaging of live cells, single-molecule force spectroscopy and recognition imaging) and combining these nanotools with fluorescence, genetics and cell biology approaches to address the cell surface.
